MaxSignal WSSV Viral Detection kit

White Spot Syndrome Virus (WSSV) is a serious viral pathogen that affects cultivated shrimp worldwide. The BIOO Scientific MaxSignalTM Shrimp WSSV Detection Kit uses an end-point Polymerase Chain Reaction (PCR) assay to screen for WSSV in fresh or frozen shrimp or other potentially infectious material. The kit contains 2 pairs of oligonucleotide primers targeting nested regions (483 bp and 388 bp) of the WSSV VP466 nucleocapsid gene. These WSSV amplicons have no significant homology to any other viral sequence. The detection limit for the synthetic WSSV Positive Control Template included in the kit is approximately 50 copies using the standard 1-step PCR protocol with 35 cycles of PCR, and approximately 5 copies using the 2-step nested PCR protocol (25 cycles for each PCR step). For detection of WSSV viral sequence in infected shrimp, positive signals have been obtained using the 1-step PCR protocol with input amounts of approximately 50 femtograms of total nucleic acid extracted from shrimp that were experimentally infected with high levels of the WSSV virus. The Shrimp Positive Control primers included in the kit are used to demonstrate successful extraction of DNA that is able to support PCR. The Shrimp Positive Control primers were designed to produce a 249 bp amplicon of the actin 1 gene from Penaeus monodon, Penaeus vannamei, and also from wild-caught crawfish. Input sample for using the kit is total DNA or total nucleic acid extracted from solid tissue or derived from liquid tissues such as hemolymph purification of viral particles for analysis is not required. The kit contains reagents for running 50 PCRs of 50 uL each the number of reactions per kit may be extended by reducing the reaction volumes. To maximize flexibility in use of the kit, sufficient PCR primers are provided to run at least 50 reactions with each of the 3 primer pairs (outer and nested WSSV primers and shrimp positive control primers). Gel Loading Solution and Molecular Weight Markers are also included in the kit. The PCR primers and other kit components are also available as stand-alone products for users who would like to evaluate any of the kit components for use with alternative assays they may currently be running.
